It is possible to turn this question on its head and make something negative into something positive. For example, you could admit that you sometimes get easily carried away with things, but that's because you love to get involved with projects and contribute as much as you can. Or you might have the tendency to be blunt with people, but you hope you're never actually rude and at least people always know where they stand with you. Etc etc.
Basically, think of a genuine fault, but think of how this 'fault' can also be helpful, and give this in your answer too.

Say something you find difficult but suggest how you deal with it. Companies are looking for solutions not problems.
eg
I do get angry and frustrated when something doesn't go my way. However I have learnt strategies to deal with this for example in my last job we had a courtyard area where I would walk around for 5 minutes to calm down and come back with a positive action.
I think the classic answer is to choose a past mistake and then show how you overcame it. I say that I used to be very nervous at presentations, but I then went on a public speaking course and now enjoy them. I like this one 'cause (i) it's true and (ii) it shows I'm willing to make an effort to overcome my problems.
