I am flying to Mumbai on a Boeing 747 from Heathrow Terminal 5 and would like to use Seatguru.com to choose a good seat. However, seatguru says gives the option of the 747-400 52-Bus and 747-400 70-Bus, both of which have different seat layouts.

Does anyone know the difference between the two, or even better, which one I will be flying on?

the difference is in the name: 52 bus has 52 business (club class) seats, 70 has 70. The only way to find out would be to ring the airline, but even then they might change the aircraft at the last moment.
PS, I suppose if it's from T5 you're on BA. In that case remember you can check in online 24 hours before takeoff and allot yourself any seat that isn't already taken by someone else doing the same thing. It means you don't have to turn up at Heathrow the usual three hours before, which has to be good.
With BA, there's every chance it could be changed for a 777 as well especially if they haven't sold enough seats to fill a 747.
