As a quick guess, I would think you have an air lock between upper and lower stories, but check pump is running first (quiet whirring .. stick a screwdriver blade to the body when running and hold to your ear) Sticking or faulty pump could cause this effect.
Is the whole system piped in copper? ... 15mm? ... or very small microbore .. 10mm or 8mm? If microbore downstairs, there may be an air lock in a manifold under floorboard .. sometimes they have a bleed and are accessible.
You "could" try turning off all upstairs rads and open one downstairs and try to bleed that first. Then others.
