I have downloaded this game for my R4 card from three different sites and each time when I go to play it it comes up with a black screen that says it couldn't read the file and to reinsert the card. Anyone got any ideas? Other games that I have downloaded have worked fine it is just this one.

Chances are, you'll need to update the firmware for the R4 card.
I had the exact same problem with a previous card i had (M3DS Simply), where most games would work fine, but newer ones came up with that message.
Unfortunately for me, the company who make the M3DS Simply, stopped doing updates for it, so i had to purchase a new card - i bought the CycloDS Revolution....a much better card all round - i'd probably advise most folks to get this one.

Try updating the R4 card's firmware first.
Failing that, i'd advise you to go for a CycloDS...

try removing a couple of other games from your card it may need a bit more space to create a save file this worked for me
