If it is T & G wooden floor boards then you need only nail it down using proper floor board nails ( cut nails ). But if it is the T & G chipboard flooring then yes to do a proper job glue the top of the joists and the T & G joints aswell plus screw them down along the joists every 300mm. If you do not you will forever have squeeky / creaking floors every time you walk on them.
In addition to gasman's always good advice, if this is a first timer for you you'll find the job will go faster, easier and better if you can rent one of these:

Here, in the U.S., they usually rent for about $75 (US) a day plus the coil nails, which aren't very expensive. You wil need a source of compressed air and small but efficient compressors will rent for about an additional $40 per day. This way, one can drive the nail with one stroke through the fist subfloor into the next which will help assure the dreaded squeak...
Best of luck!

Firstly, if you are using timber t & g boards lay stretches of 5 boards and then cramp them together using floorboard cramps and then nailing them along the edge of each board with 3" losthead nails. Use a thick plank between the cramps and the boards to prevent damaging them.
Secondly, if you are using chipboard floor panels stagger the joints and screw them to the joists using 2" screws but I would not recommend gluing them to the joists or to each other because if plumbers or electricians need access at a later date you will regret it.
