Well in the sense that the insurer would use it as an excuse to not pay out in the event of a claim. Your insurance is still valid, you would not be prosecuted for no insurance.
Insurance companies are obliged by law to honour any third party claims irrespective of whether your car is MOT'd.

However, where claims are made in these circumstances the companies can (and do) sue the insured for the cost of the claim.
If you have no MOT then you have no proof that the car is roadworthy. This could invalidate your insurance if you have an accident as they will assume your car failed and should not be used.
