Yes most recently in Lethal Weapon they killed off Martin Riggs for season three... he doesn't die in the films played by Mel Gibson.

Don't get me started on 'Game of thrones'.

Joss Whedon habit of killing big characters is almost a trope now.

Buffy: Tara, Anya, Spike (later ressurected though)
Angel: Doyle, Cordy, Fred, Wesley
Firefly/Serenity: Wash, Shepherd Book.

I was going to mention 'Supernatural' but you may not have seen it.... a lot of leading characters have being and gone from that.
