I have a mini to sell, worth about £6,000, and I want to buy a car for £4,000 that I will get more mileage to the gallon. Also, as I sell my car, I will need a car to use straight away for work. What is the best way please?

Since you do not want to be without a car (for any period), your only options are to either part-exchange the car or buy before you sell.
You could try advertising your Mini and stating that a cheaper, more economical car, could be taken as part payment. However, unless you go for a car with a much smaller engine [although you might consider diesel] will the change be worth it? After all, Mini's are quite good on fuel and if your driving style and journeys reflect poor mpg then would a replacement show significant improvement?
If your bank will give you an overdraft of 4k use that to buy the car and pay it off in full as soon as you sell your car. The interest shouldn't be too much if it is paid off within a month or two.
Or if you buy from a garage, see how much they are prepared to give you for your car.
You will have a job getting a car which does many more miles per gallon than the mini.
Go to a used car dealer and tell them you want to sell the Mini and buy a cheaper car, they will probably do a deal, Minis are in demand so they will be able to resell at a profit.
Otherwise get a cash buyer for the Mini and when you have cash in your pocket go out and buy another car.
For more mpg figures have a look at www.honestjohn.co.uk and check out the Real MPG section. Sorry to go off subject a bit but I'm just trying to help prevent you changing your car "for something more economical" when what you might lose on your current car will more than offset a slight gain in mpg.
You can buy a lot of fuel for that kind of money and you're buying a car of unknown reliability. I would think the resale value of the Mini would remain high and serve you better in the end.
It also occured to me your title and tags will use some of that up.
how would you like a ford focus tdci value approx 4500 2007 plate
