Lady Marmalade wasn't written for this film. It was a hit in 1974 for Labelle (Patti Labelle's group) and written before that but wasn't a hit.

I think it was a bit of a vehicle for Pink, Mya and Christine A-Guerilla (yes thats on purpose) to get them out there and Missy Elliot and Lil Kim were known and established artists.

Some of the The lyrics :

Hey Sister, Go Sister, Soul Sister, Go Sister
Hey Sister, Go Sister, Soul Sister, Go Sister

He met Marmalade down in Old New Orleans
Struttin' her stuff on the street
She said "Hello,
Hey Joe, you wanna give it a go?" Mmm, Mmm.

Itchi Gitchi Ya Ya Da Da
Itchi Gitchi Ya Ya Here
Mocha-choca-lata Ya Ya
Creole Lady Marmalade

Voulez-vous coucher avec moi ce soir?
Voulez-vous coucher avec moi?

He sat in her boudoir while she freshened up
The boy drank all her magnolia-wine
On the black satin sheets where he started to freak
Hey, Hey, Hey
Touching her skin feelin' silky smooth
The colour of cafe au lait
Made the savage beast inside
Roar until it cried, More, More, More

Now he's back home doing 9 to 5
Living his grey flannel life
But when he turns off to sleep
Old memories creep, More, More, More

Voulez-vous coucher avec moi ce soir?
Voulez-vous coucher avec moi?
Voulez-vous couchez avec moi ce soir?
Voulez-vous coucher avec moi?
Mmm, Hmmm

Itchi Gitchi Ya Ya Da Da
Itchi Gitchi Ya Ya Here
Mocha-choco-lata Ya Ya

So she was a prostitute in Old New orleans !
think you thought the song featured in the film because of the outfits they wore in the video, very moulin rouge-ish :-)
Well the film is set in a brothel, so the song and its lyrics (which translate as 'do you want to sleep with me tonight?) fit pretty well in the context..
The song Lady Marmalade is on the soundtrack for the film so it must have been to do with the whole prostitute thing.
The song is used at the beginning of the film (10:50) when they first go to the moulin rouge.... If you've got it on DVD its the chapter titled "at the moulin rouge you'll have fun" and its remixed with nirvanna smells like teen spirit and the can-can
