Peroxidase staining detects myeloperoxidase present in the primary granules of myeloid cells. Myeloperoxidase is present is virtually all immature and mature myeloid cells and in some other cells such as monocytes.

Myeloperoxide in the presence of H2O2 releases oxygen that can be detected by a number of reagants including 3-amino 9-ethyl carbazole, which turns brown in colour and benzidine, which turns black in colour.
