Force delete of files

There is some annoying spyware on my computer.
System32 folder:
jkkji.dll
urqopmm.dll

I can't delete them because it keeps saying that they are in use.
I can't get into safe mode, i've tried 3 virus scanners and removed them but they keep coming back, i've tried killing the process, it doesn't work, i've tried deleting them from the registry they keep coming back, i've tried using a program to forcefully delete them, they still come back on reboot, i've tried unregistering the dlls but i get an error message that the dll was loaded but could not be registered.
I'm at the point of throwing the computer out of the window!
Any help welcome.

Hi Wix,
Have you tried running the virus scanner from the command prompt rather than Windows? It could be worth a try.
If not you may be able to go back to the time before your pc was infected using system restore. If that fails then try to restore from a recent backup disc( do you do make regular backups? )
Failing that then use the system reload disc that puts your machine back to how it was when it left the shop / factory. That will most certainly kill off any spyware/ virus problems but it is a last chance option . Please note that any personal files / pictures / music / videos or progams saved to your hard drive will be deleted by doing this. Good luck.

Thanks to all of you for your advice.
I ended up deleting the registry keys, blocking the process and i disabled my internet from using 3rd party BHOs.
Its all worked, the virus file is still on my computer but can't do anything.

Will keep instructions handy in case of re-infection.

UPDATE:
jkkji.dll file was damaged, got rid of it with some related hidden ini files using ComboFix.
Got rid of urqopmm.dll using some ant-spyware software.
