First thing is to sample the atmosphere of both Kuta and then Senur Beach. This is where you do do some serious partying, eating and beach/pool stuff. Be prepared both are very busy places and often do not meet images of Bali people have in their heads. Once your ready to move on head upto Ubud about and hours drive north. Great place where you do the same things as Kuta and Senur but a little more relaxed pace, not so busy and alot more cultural. Some great hotels and spa's here. The monkey forest is here but not the most rewarding wildlife experience! Once you are ready to move again head east to the temple of Besakik next to the impressive Gunung Agung volcano (can be climbed if your fit). Move from here to east coast (Candi Dada or Culik) for more relax beach stuff and good snorkelling/diving. Travel round the coast north to Singaraja for peace and quiet, some good hot springs near here. Travelling south again the best route is past Gunung Batur volcano (trips up for sunrise easily organised) and via Tampaksiring. All Bali hotels are cheap and easily booked at tourist agents dotted all over the island as to is transport. All the buses are frequently used by travellers and safe. You can do good deals with taxi/bemo's if you prefer. Example, bus Denpasar to Ubud 1.80, taxi 7. Do you shopping last off, you could buy alot of stuff, ideally in Ubud or Denpasar instead of Kuta which has higher prices. If you want to go further you can ferry or fly to Lombok very cheaply and with very short notice. From here you can also get to the Gili islands which are worth a visit or short stay. Finally bargin hard deals can be done on lots of things while your there, especially accomodation which is still suffering badly from low occupancy after the bombings. ENJOY!
