When a hotel asks for my credit card number to guarantee a booking, do they take the money for the room there and then or is it just to guarantee the booking?

They take the money when you pay your bill.
A lot take your credit card details so if you don't turn up on first night without cancelling they can debit your card for that night. It's just an insurance for them. Some do however, take say a 10% deposit which isn't refundable before say a month of your visit but this must be stated at the reservation.
