You can have your girl scanned at 4 weeks onwards and they can tell for sure, and give an approximate idea of how many puppies to expect.
Other signs, some go off their food, others have morning sickness which is usually just bile and others show no signs until 4 weeks then go off food, they may also be quite clingy. Some have blue rings around teets at 3 weeks, and a creamy discharge at around 6weeks. (discharge must not be dark in colour). Some show no signs at all, and you dont know until they go into whelp.
Once you know for sure, you can increase food and protein intake, as the puppies will be taking most of the mothers nourishment. Some breeders give the in whelp females puppy diet, and use the same diet to wean the puppies. If she is in whelp, dont forget to give your girl calcium tablets to avoid a calcium drop which can be life threatening.

On the calcium, I would say do not give this until the puppies are actually born because supplementing the calcium can cause problems. They do not need it until the pups are taking milk from the bitch and will drain her of calcium. Not supplementing at that time can cause eclampsia which can be fatal if left untreated.
