When booking a holiday, the travel agent says something like £344 based on 2 adults sharing. Does that mean it costs £344 per person, or £344 for both?

It's not obvious but I would guess it means £344 each. If it was based on 4 sharing for example it might well be cheaper eg £310 each.
each. The idea is that if you're travelling alone, you'll be paying more than £344 because they'll hit you with a single supplement (generally based on the notion that one person takes up just as much space in a hotel as two people).
The price quoted of £344 is per person based on 2 people sharing a room. If you were to go on your own you would pay twice that.
