Substitution

What is the position if play has been stopped and the referee is directed to a substitution that is being made (the player having been injured). The fouth official has held up the board showing the subsitution. As he is walking off, but still on the pitch, the player shouts to his manager that he is okay and the manager changes his mind about the substitution and and tells the proposed subsitute to sit back down. Is this allowed?
