File won't save changes. Can you help.

I have an Excel membership file on a flash drive that I've used for a few years now. I made a few changes to it this morning, which I do regularly, but it won't save the changes and I don't understand the reasons it's giving, which are:
1. The file name or path doesn't exist. (It must exist or it wouldn't open)
2. The file you're trying to open is being used by another program. Close the document in the other program and try again. (it isn't being used by another program).
3 The name of the workbook you're trying to save is the name as another document that is read only. Try saving the workbook with a different name. ( I have but I still get all 3 of the above)

Won't save any changes to any files and documents on the flash drive. I've transferred everything to my external HD and intend to delete everything from the flash drive then transfer it back again.
