Are there any writers, artists etc who failed during their lifetime and, more importantly, gave up hope as a result? (And who had their works published posthumously?

Check out the Bronte sisters - apart from Charlotte, not successsful during their short lifetimes.

John Kennedy Toole wrote A Confederacy of Dunces which was a great success. Sadly, after his death by suicide, I think.

Not that I'm aware of. They died at early ages from common maladies.
