The guy is probably just teasing you, and he KNOWS your butt is not big. It's probably because you make an issue of it that he even noticed it ( running morning AND night seems a bit much..). My Brother used to tease me about my nose all the time because I kept saying I thought it was too big. It is not. But he just liked to tease me about it because he knew I cared. It's probably just good fun for him, don't go calling him a "fat *******" or anything insulting ! On the other hand, if he starts groping you or the comments become too much, and definitely sexist, then I guess a good old slap is "de rigueur" ! ;-) Good luck.
I know you're not a lardy bum like him but...you could say "Well you're ugly but I can lose weight".
Or alternatively, rather more to the point, "Can it fat boy!!" This comment may get YOU into trouble though for being 'fat-ist'. A forceful yet calm "F*** off' might do the trick and prevent you from being disciplined for size-ist remarks.
But seriously, a personal comment like that from him just isn't acceptable!!!! Absolutely not!. Years ago he may have got away with it but not in these enlightened times. I'd consider mentioning it to your line-manager or Head of Dept. If these are likely to be tricky because your manager is equally 'blokey' or a friend of your colleague, then seek out a supportive female in Personnel/Human Resources.
It is in no way okay to say anything like this to a colleague, male or female and he needs to be stopped.

Just turn round to him seriously and say "Okay, you have made your point, as offensive as it is. If you mention my butt EVER again I am going to get a disciplinary against you for harassment. Do you understand?". Then wait for him to reply. When he admits he has understood, drop it. If he continues to make comments, go to your boss and explain how this is upsetting you and affecting your work. Your boss should then be able to sort it for you.
charisma - I really believe that it is this chap's warped sense of humour.
People never tease about real problems - if you were fat he'd tease you about your beautiful hair if he is bald, for example.
It's a weird way of poking fun at himself.
Just tell him to stop, or ignore it.
If he is looking at your backside in a pervy way, tell him if he doesn't stop you will report him for sexual harrassment.
Good luck.
