where does it come from (ie where is the studio based - is it in manchester like the rest of r5 or is it london so simon can do his radio 2 show from london afterwards?

Simon Mayo left 5 Live when they moved to Salford, so he could stay down south.

As you say, he has a Radio 2 show afterwards so it must all be done in London.

Mark Kermode also records a film review article for BBC News Channel, and they too are in London.
