How does one determine the root note of a chord?

presuming you mean for guitar, the root note to a chord is through the base note.
By 'base', sheffwed means 'bass', ie the lowest note, so for any E chord, for example, the lowest note is the open E string. I'm assuming it's similar for other instruments.
