Should I Contact Human Resources?

I was hired last October at a beauty store as a licensed esthetician. Upon interview I told my possible employer that I can only work two and a half days a week because my full time job is actually my religious activity. She said that was fine and proceeded to set my schedule that day. A few months down the road that same store manager as well as my esthetician manager pressured me to work a fourth day, full shift. At first I said no I can't but they were practically begging me saying we're understaffed, we really need you, yada yada yada. After about two weeks of that I agreed because I assumed it was only temporary until they hired another esthetician (there was 3 of us at the time). So a few months ago I went up to my retail manager who is my main manager and I asked her if she can give me my original schedule back so I can go back to my full time missionary work. She said that, “that doesn't work for us and the company so no. Besides, on your resume you put open schedule so there's really nothing I can do…”
So now I'm kicking myself because I put open schedule but I only did that because I knew no one would want to hire someone who will only work 2 1/2 days. I wish during the first interview I had her sign something agreeing to those 3 days.
Present time we now have a fourth esthetician and I'm planning on asking my manager again if she can give me my original schedule back and if she says no again I feel it's necessary to contact human resources at that point because I did not agree to this upon hire and it greatly conflicts with my religious schedule
