last week at Brighton Pavilion, i was listening to the audio tape and i am sure it said " Within 3 years, he (GIV) lost Charlotte, his wife and his daughter and his eldest brother".
How could he have an eldest brother? Surely that brother would have been the king?

Well, in answer to your question, he didn't have an older brother.
His wife was Caroline of Brunswick - who he hated with a vengence - he famously got so drunk on his wedding night that he passed out in the fire grate, he barred her from his corronation I doubt he'd have mourned her passing in 1821.

He did have a daughter Charlotte by her though - she died in 1817 aged 23

I think they were talking about William IV

George IV was Williams older brother (d. 1830). as was Frederick (d. 1827). He had 2 daughers and a son all of whom died in infancy 1819, 1819 and 1822 and was married to a Charlotte (Augusta Louisa) who died in 1819
The eldest brother isn't always King, they can die before their Father, as in the case of Henry VIIIs older brother Arthur. Henry married Arthurs widow, Catherine of Aragon and this was the basis for his divorce from her many years later.
