If you look in this blooper, there is a scene very near the start where Will, Grace, Jack & Karen are all sitting in a car but the background is blue (like out of the rear window and other windows). Is this because they don't really drive but they then edit it to make it look like they are in manhatten or so on? Also what do they find so funny in that scene, I can't work it out.

It is spelt Chromakey and is most often blue or green (sometimes referred to as "green screen"), but can be red or...well almost any colour these days. ( the TV picture is made up of red, green and blue points of light).
The background inserted into the picture will most likely be film or video tape recorded earlier or more than likely stock footage (TV studios build up a library of such footage).
All done because it is cheaper than mounting the car on a low-loader (a low trailer with the car on it and the camera mounted nearby), or bolting a camera to the car and having one of the actors drive and deliver their lines.
Either way the footage would have to be recorded in advance, maybe on a day when the artists would be better rehearsing that week's show, so to mount the whole thing in the studio is easier.
