For more on marking an answer as the "Best Answer", please visit our FAQ.Flash Player has a nasty habit of not working, even though it remains installed. When you try to reinstall it the process is reported as successful but the old (non-working) files seem to take precedence over the newer ones (so Flash Player still refuses to work).
Go to Add/Remove Programs. Uninstall Adobe Flash Player Plugin, reboot your PC and then try a fresh installation.

Here's where (the equivalent to) Add/Remove Programs is hiding in Vista:

If uninstalling, and then reinstalling, doesn't work it might be necessary to try a repair installation of Vista:

On my ancient PC (running Windows ME and using Firefix as my browser) it shows as 'Adobe Flash Player Plugin'.
Mentioning browsers reminds me that it might be worth trying a different browser. (i.e. the problem might be with the way that Flash Player works, or doesn't work, with your existing browser). Firefox is vastly better than Internet Explorer:

Some people prefer Chrome:
