If I charged my mobile phone from my cigarette lighter socket without running the engine, would it flatten the battery? It's an iPhone 6 and the vehicle is a Vauxhall combo van.

Not all cigarette lighter sockets are live when the ignition is off, so you may not be able to.
If it is live there will be no problem - I frequently charge cellphones, camera batteries etc using a socket which is live with the ignition off.

it would eventually but you should be ok for once in between engine starts, though often they are not live with the ignition off.
If it can start a diesel engine, then I doubt an iPhone will trouble it.
I have a Suzuki ignis and if i use the plug in car vacuum from the cig lighter it flattens the battery and after that unfortunate error and the tutting from the AA man I don't risk using the cig lighter thing for anything now.
Car vacuum cleaners take around 10 amps, phone chargers take milliamps. It's the current draw which flattens the battery.
If your battery was on its way out, then, yes it might. Best to wait until you are on the road, so to speak.
